Remind yourself what the Ternary Complex Model (TCM) (See uploaded Reading-TCM essay source paper) is from the lectures and then read the annotated version of the *paper posted in this folder on the VLE. This annotated version has comments added (see highlighted yellow text) which will help you to avoid getting bogged down in minutiae of the protocols and maths, and will help you to focus on the pharmacological data. Your concise essay should include sections on
1. Introduction. Very brief summary of the background and the group’s previous paper (ref 6.). What was the “mutant receptor” used throughout the Samama paper and how did they come about making it? What did Samama et al set out to address?
2. Pharmacological data. What were the pharmacological differences between the wild type and the mutant receptors? Describe these clearly and concisely, demonstrating that you understand the outputs – include figures from the paper if it helps you
3. Discussion. Include the following within your discussion. What aspects of the pharmacology of the mutant receptor can the TCM explain? Why could these data not be explained by simpler models (Clark/Operational model)? What data in the paper cannot be explained by the TCM (and why)? You do NOT need to explain the extended TCM – you only need to explain why the original TCM was sufficient to explain most, but not all, of pharmacological data.
